Abstract

Epigenetic control mechanisms for gene expression are paramount in the differentiation and maintenance of the characteristics of stem cells. The REST transcription factor is essential for keeping the expression of neuronal genes in non-neuronal tissues repressed through various epigenetic mechanisms, as well as for the maintenance of pluripotency and self-renewal in stem cells. The BHC multiprotein complex is a REST co repressor through the action of histone deacetylases and histone H3 demethylase at lysine 4. This project investigates how the activity of the BHC complex is regulated by the interaction of two homologous proteins called BRAF35 and iBRAF and what are the consequences of this regulation in neuronal differentiation and pluripotency.
