Personalized therapies in ovarian cancer to overcome resistance

Abstract

Ovarian cancer (OC) is the gynecological cancer with the highest mortality rate, largely due to high resistance to conventional therapies. Therefore, finding more effective treatments overcoming chemotherapy resistance is of vital importance and a priority in cancer research. OC is characterized by high rates of genome instability, which is likely caused by DNA repair defects, and is treated with platinum-based compounds, which are genotoxic agents, in the first line. Although new therapies targeting DNA repair, as poly-ADP ribose polymerase inhibitors, have improved patient outcomes in relevant phenotypes, many patients will not benefit from these therapies and therefore a better stratification is required. In this project, we will search for new DNA repair therapeutic targets that can overcome chemotherapy resistance in OC. The results of this project have the potential to aid in personalizing treatments of OC patients based on predictive molecular biomarkers, generating greater efficacy and less toxicity. Consequently, our project has a direct impact on the clinical management of OC patients and will contribute to improve the survival and quality of life of these women.
